Rab2 and Rab6 are Implicated in Acrosome Formation during Spermatogenesis in Eriocheir sinensis: Based on Sperm Proteome

Background: Rab proteins are GTP-dependent small that function as regulators of intracellular vesicle transport, fusion, and localization. However, few studies have investigated their in Decapoda reproduction. The Eriocheir sinensis sperm has no tail the nuclei uncondensed. With acrosome forming majority mass, it provides an ideal model for studying formation. Methods: We firstly analyzed proteome using LC-MS/MS. To study functions Rab2 Rab6, related to Golgi apparatus, formation during spermatogenesis, genes Rab6 were cloned based on testis transcriptome E.sinensis poly-clonal antibodies prepared. presence 2 was confirmed by western blot. further observed characteristics target immunofluorescence (IF). Results: A total 1247 including 7 proteins, Rab1, Rab2, Rab5, Rab11, Rab14, Rab18 identified proteome. IF results showed co-localizes with GM130, a cis-Golgi matrix protein, spermatagonia spermatocytes. In early spermatids, participate pre-acrosomal vesicles. maturing both settle acrosomal membrane but present different wrapping pre-acrosome. mature sperm, localizes perinuclear theca surrounding cup, while remains membrane. Conclusions: Our research found analysis E.sinensis, involvement These findings provide foundation spermatogenesis animals.
